Efficient pre‑treatment for pancreatic cancer using chloroquine‑loaded nanoparticles targeting pancreatic stellate cells

چکیده

Pancreatic stellate cells (PSCs) play a key role in desmoplastic stroma, which is characteristic of pancreatic ductal adenocarcinoma (PDAC), and they also enhance the malignancy cancer cells. Our previous study reported chloroquine's mitigating effects on PSC activation; however, drug known to induce adverse clinical practice. The present aimed reduce chloroquine doses develop useful pre‑treatment that targets PSCs using nanoparticles. Poly lactic‑co‑glycolic acid (PLGA) nanoparticles were used as carriers loaded with indocyanine green (Nano‑ICG) or (Nano‑CQ). Tumor accumulation Nano‑ICG was evaluated an vivo imaging system. chloroquine, Nano‑CQ and/or chemotherapy gemcitabine investigated orthotopic xenograft mouse model. selectively accumulated tumors persisted therein for over 7 days after administration. Additionally, peritoneal metastasized regions, but not liver, kidney normal tissues. reduced density activated at lower significantly restrained tumor progression combination gemcitabine. In conclusion, PLGA nanosystem successfully delivered tumors. efficiently activation may be promising novel strategy PDAC.8

Pancreatic cancer-associated stellate cells

Pancreatic cancer-associated stellate cells secrete soluble factors, such as interleukin-6 (IL-6), that promote the accumulation of myeloid-derived suppressor cells via a signal transducer and activator of transcription 3 (STAT3)-dependent mechanism. Targeting components of the IL-6/JAK/STAT3 signaling axis within the tumor stroma could therefore inhibit local immunosuppression and improve the ...

Pancreatic stellate cells and pancreatic cancer cells: an unholy alliance.

Pancreatic cancer--a tumor displaying a particularly abundant stromal reaction--is notorious for its poor prognosis. Recent studies, via newly developed orthotopic models, provide compelling evidence of an important role for pancreatic stellate cells (PSC) in pancreatic cancer progression. Characterization of the mechanisms mediating PSC-cancer interactions will lead to the development of much ...

Pancreatic stellate cells: partners in crime with pancreatic cancer cells.

Pancreatic stellate cells (PSC) produce the stromal reaction in pancreatic cancer, but their role in cancer progression is not fully elucidated. We examined the influence of PSCs on pancreatic cancer growth using (a) an orthotopic model of pancreatic cancer and (b) cultured human PSCs (hPSC) and human pancreatic cancer cell lines MiaPaCa-2 and Panc-1. Athymic mice received an intrapancreatic in...

Role of pancreatic stellate cells in chemoresistance in pancreatic cancer

Pancreatic cancer is highly chemoresistant. A major contributing factor is the characteristic extensive stromal or fibrotic reaction, which comprises up to 90% of the tumor volume. Over the last decade there has been intensive research into the role of the pro-fibrogenic pancreatic stellate cells (PSCs) and their interaction with pancreatic cancer cells. As a result of the significant alteratio...

Desmoplastic reaction in pancreatic cancer: role of pancreatic stellate cells.

OBJECTIVES Pancreatic cancer has a very poor prognosis, largely due to its propensity for early local and distant spread. Histopathologically, most pancreatic cancers are characterized by a prominent stromal/fibrous reaction in and around tumor tissue. The aims of this study were to determine whether (1) the cells responsible for the formation of the stromal reaction in human pancreatic cancers...
